Chicken insulin like growth factor Ⅰ(IGF?Ⅰ)labeled by 125 I was incubated with IGF?Ⅰ degradation enzyme(crude extraction from kidney)under various conditions,such as incubation time,temperature,pH,and content of crude enzyme.
结果显示 :①IGF Ⅰ分解酶反应时间以 30min为宜 ;②粗酶液的含量宜在 1 0 8μg·μL-1以下 ;③酶活性在 4 5℃时最高 ;④此酶在 5 0℃以下稳定 ,当温度升至 5 5℃时其活性急剧下降 ,升至 6 0℃时将降至最大活性的 2 0 %左右 ;⑤此酶最适pH值为 6 5~ 7 0。
